Not exactly.

A portion of the PA Turnpike is built on the grade of the "South Penn" - the South Pennsylvania Railroad:

EBT was used to haul materials for building the Turnpike to Shade Gap, where it was hauled to the Fort Littleton area. There were lots of plans for laying additional trackage, including a branch from Shade Gap to Fort Littleton, but none came to fruition.

Rainey and Kyper's book is the best source for this information.
