I don't know about speed, but wheel size definitely affects the loading structure. A larger wheel size has a larger footprint on the rail, thus allowing the cars and the loads to be heavier. They can also allow a larger axle, and bearing surfaces which also increases the capacity.

Today's SG freight car wheels are significantly larger than the 33" wheel standard of the past for these reasons.
